Effects of Everolimus on oxidant-antioxidant system and on hepathic histology in experimental hepathic -ischemia reperfusion injury

Abstract (EN)

Ischemia-reperfusion (I/R) injury is one of the mot common ethyology of hepatocellular damage occure after liver surgery. Achievement of transplantation surgery is depends on I/R injury during operation. This issue can be resulted as fulminant hepatic failure, acute-chronic rejection, early retransplantaion and death by a ratio of 10-20 %. So, recognizing and preventing the mechanisms of hepatic I/R injury, is important for increasing the success of transplantation surgery.In this study, we investigate the protective effect of everolimus in hepatic I/R injury which is an macrolide antibiotic and also an immunsupressive drug for heart and kidney transplantation by mTOR inhibition.We used 24 female Winstar albino rat with a weight of 200-250 gr. The animals were randomized into four groups. Group I; sham (control) group whom didn?t given medical treatment, only laparotomy was performed and blood and liver tissue samples were collected. Group II; I/R group whom also didn?t?t given medical treatment. An atravmatic clip was used to prevent blood supply to the left lateral and median lobes of liver. After 60 minutes of hepatic ischemia, the clip was removed to recover hepatic reperfusion for 24 hours and blood and liver tissue samples were collected. Group III; I/R group whom were treated with 1.5 mg/kg/day everolimus peroral without performing ischemia. After 7 days blood and liver tissue samples were collected. Group IV; everolimus + I/R (treatment) group whom were treated with 1.5 mg/kg/day peroral everolimus .After 7 days of treatment, ischemia were performed as explained above and days blood and liver tissue samples were collected.To show the efficiency of treatment AST, ALT, albumin, IL-6, TNF-?, tissue MDA and SOD levels were investigated. Histopathological investigation was performed from tissue samples.In conclusion; athough in histopathological investigation, establishing intraparanchimal necrosis in three rats from treatment group is challenging, istatistical significant recovery in biochemical results supports the efficiency of everolimus in hepatic I/R injury.
