Pregabalin during acute phase of experimental head trauma, anti-edema, anti-inflammatory and protective effect

Abstract (EN)
Objective: Traumatic brain injury, includes primary and secondary mechanisms of injury. Secondary damage, after hours or days, depending on the emerging and is composed of many complex pathophysiological mechanism. In the studies, pregabalin with ?2-? subunit of calcium channels, strongly connected, the result of glutamate, noradrenaline, serotonin, dopamine and substance P causes a decrease in the release of various excitatory neurotransmitters. In our study, rats were created severe closed diffuse brain injury, trauma after the creation of pregabalin in the treatment of acute given antiödem, anti-inflammatory and neuroprotective effect was investigated.Methods: This experimental animal study, the weights from 300 to 350 g adult male Sprague-Dawley rats ranging from 32 units were used. Subjects were eight rats in each group so that the control (K), trauma (T); trauma with pregabalin (T + P); only pregabalin (P) groups. Pregabalin study, diluted with saline intraperitoneally. Medication after trauma 84 h of the rats were decapitated for examples histopathological examination.Results: The group with trauma after trauma there is no difference in bleeding between the pregabalin-treated group (p = 0,368), no significant difference in terms of inflammation (p = 0.003) was found. Again, no statistically significant difference between these groups there is edema (p = 0.131). Trauma group, 8 subjects, 6 subjects in the group post-traumatic edema was pregabalin. Edema was observed 2 subjects.Conclusion: The results of our study in acute traumatic brain injury, traumatic brain injury secondary to pregabalin treatment in reducing inflammation may be protective against the show. As a result of pregabalin in the treatment of humans with acute traumatic brain injury was thought to be a useful option.
