Abstract (EN)

Recent biotechnological research has focused on the synergistic integration of different approaches to maximize the benefits of a process. The enzyme industry is a clear example of this strategy, and the infinite possibilities that are being evaluated to establish more efficient, environmentally friendly and cheaper means of production have marked the path of a new innovative era. This research investigates three important aspects that are coupled to lay the groundwork for a process at the forefront of the mentioned characteristics. The purpose of the research was the evaluation and application of the statistical methods (Taguchi methodology and the Surface Response Methodology) to optimize the production and activity of the xylanase enzyme isolated from the marine-derived strain Trichoderma pleuroticola 08ÇK001 using as substrates agricultural residues which are discarded as non-valuable materials. The results were found satisfactory in terms of the increase in xylanase activity and the optimal predicted process conditions. The optimum compounds and values were as follows: carbon source; pistachio hull (18.48 g/l), nitrogen source; tryptone (11.15 g/l), fermentation time; 24 h, salt concentration; 50 g/l, pH; 8.0, inoculum quantity; 2.5 ml and spore concentration: 5x106 spores/ml. At the end of the research, the total increment in the activity, compared to the values measured at the beginning of the experiment, was 117.2% and 314.1%. The final optimized experimental specific activity was 147.8 U/mg. The enzymatic activity and the general results of the research are promising for industrial applications, especially in the paper bleaching industry due to its alkaline nature or in the seafood sector, due to its halotolerance.
