Abstract (EN)
In this thesis, optimum production media and conditions for the production of xylanase in submerged culture fermentation has been determined. Different agricultural wastes (wheat bran, malt sprouts, corn cobs, rice straw, sawdust, oat bran) was used as substrate for production from Aspergillus niger. Concentration of carbon and nitrogen sources was determined. Maximum activity (766,16 U/ml) was obtained at 5% (w/v) malt sprout concentration. Inoculation size, aeration and stirred rate and dissolved oxygen parameters were optimized in 2 liters stirred tank bioreactor and maximum specific activity (164,70 U/mg) was measured at 30% dissolved oxygen, 0,5-1 vvm aeration rate and 96 hours of incubation period. Key words : xylanase, malt sprout, submerged cuture fermantation, Aspergillus niger
