Abstract (EN)

Biotechnology, the key technology of the 21st century, has influenced the cosmetics industry in line with the demands of more efficient and safe products. Annual growth rate of 6 % in anti-aging skin care products increased the effectiveness of bionanotechnology products in the market. In the scope of this thesis study, for the purpose of producing products against skin aging, the inhibitor peptide (PEP3) was designed for MMP-12 enzyme inhibition. PEP3 with 13 µM concentration inhibited the enzyme MMP-12 by 88%. In Western blot experiment conducted with PEP3 with 10 µg/ml concentration on the HS68 cell line, an increase was observed in the amounts of collagen I, IV, elastin and fibronectin proteins. PEP3 was encapsulated with solid lipid nanoparticles (SLN) and transdermal release study was performed. The loading efficiency was found as 59,92%. The PEP3 loaded SLN was dried with mannitol and the stability study was performed. The nanotechnology safety evaluation of the formulation of PEP3 containing SLN resulted in a wrinkle reduction of 18% compared to baseline after 28 days of use. PEP3 has been presented to the dermocosmetics market under the brand name Dervatyl™ (dermal renovation peptide).
