Possible effects of Resveratrol on damage and function disorders of Epididymis and Ductus deferens on account of Di-n Buthyl Phthalate (DBP)

Abstract (EN)
Di-butyl phthalate is a chemical that changes function and development of the endocrine system and also involved in industrial products. The antioxidant Resveratrol has very important biological effects. For all these reasons, we aimed to observe the possible protective effects of Resveratrol which is an antioxidant against the damage of an endocrine disruptor di-butyl phthalate on epididymis and deferens with the immunohistochemical, TUNEL and electronmicroscobic methots.20 days old, 36 male Wistar albino rats used.They divided into 6 groups. Each group has 6 experimental subjects. I.Group: Control, II.Group: Solvent(CMC-10ml/kg), III.Group: 500 mg/kg/day-DBP, IV.Group: 500 mg/kg/day-DBP+20mg/kg/day-Resveratrol ,V.Group: 1000 mg/kg/dayDBP, VI.Group: 1000 mg/kg/day+20mg/kg/day-Resveratrol. Finally, deferens and epididymis tissues were taken and turn to blocks through the usual methods of light and electronmicroscopy methods.Degeneration of stereocilia and head anomalies of spermium was increased parallel with increasing of DBP. C-kit immunstaining in Resvaratrol treated tissue wasn?t effective with the high doses of DBP. In deferens between groups, while a significant difference wasn?t observed in c-kit immunestaining, in the DBP-Resveratrol treated groups membranous c-kit immunreactivity increased. In TUNEL, there wasn?t recognizable apoptotic cell. 1000 mg-DBP-Resvratrol treated group was partially preserved the ultrastructure of the epididimis but insufficently. The ultrastructure was preserved by resverotrol but stereocilia degeneration occured in the high dose.In the result higher dose DBP elicits structural degeneration in epididymis and deferense, and using of resveratrol can turn these changes back by its protective effects. But in higher dose DBP, Resveratrol was insufficient. Therefore, the aim of reverting the damage that increase with increasing DBP dose, studies about resveratrol dose was considered to be necessary in Resveratrol applications.
