Abstract (EN)

In this study, it is aimed to obtain DNA sequence from Docotettix cornutus adults collected from pomegranate orchards and barcode them. Samples were collected from pomegranate orchards in Aydın, İzmir and Manisa provinces using japanese umbrellas. The first species identification of Docotettix cornutus was made, and DNA isolation processes were carried out. Sequence analysis of our samples was performed using HLO and HCO primers. When the intragroup DNA differences were examined, healthy readings were found between 499bp (SE05) and 675bp (SE07). In our samples, Thymine was found to be %33,33 - %33,89, Cytosine %14,19 - %15,03, Adenine %31,86 - %32,94 and Guanine %18.81 - %20.24.When the distance between samples within the group was calculated, it was determined that SE05 and SE07 carried the same haplotype, while SE15 and SE5 samples contained the haplotypes farthest from each other. In the analysis made with the BlastN sequence analysis program, it was determined that the closest species to Docotettix cornutus is Synophropsis lauri.DNA sequences in the COI gene region of each sample used in the study were determined. DNA sequences were compared with samples that could be similar on the NCBI base by BLAST. The NCBI code number was taken for each sample and recorded.
