Abstract (EN)
Pseudomonas bacteria are widespreadly occurred in the nature. P.aeruginosa is the common isolated species in this genus. This microorganism is foremost factor for nasocomial infections. It causes serious infections when host defence is broken down. Various virulence factors have major roles in the pathogenesis of P.aeruginosa infections.Our study is carried out to search virulence factors such as; biofilm, elastase, alkali protease pigment, exotoxin A, phospholipase C and adhesion in the origin of 155 Pseudomonas organisms isolated from natural and clinical samples.In our research totally 104 clinical Pseudomonas isolates are measured for 24 hours. Biofilm is found strongly gram positive in 56 of isolates ((%53.84), in the study that is done by using crystal violet. In the study that is carried out by safranin, it is found gram positive in 65 (%62.50) of isolates and it is found that 50.98 is gram positive in 56 nature samples totally.In 48 hour measurement, it is found strongly gram positive in isolates where, crystal violet, and safranin used studies and totally 51 nature samples are found as 42 (%40.38), 50 (%48.07) and 21 (%41.17) respectively.In our study, elastase activity in 104 clinical Pseudomonas isolates is found positive in 82 strains (%78.84). It is found positive in 38 of 51 natural isolates. (%74.50). Alkali protease activity is found positive in 74 (%71.15), of clinical isolates and 29 of 51 (%56.86) natural isolates.In the 104 clinical Pseudomonas isolates, phospholipase C activity is found positive in 74 of 104(% 71.15), and 34 of 52 (% 66.66) natural samples according to method 1.According to method 2, 66 of 104 (% 63.46) clinical Pseudomonas isolates are found positive. On the other hand 33 of 51 (% 64.70) natural isolates are found positive.In our study, pyocyanin production is found, in 35 of 100 P. aeruginosa clinic isolates while pyoverdin is found in 63 of 100 isolates. Fluorescein is occurred in 1 of 3 P.fluorescens isolates. Exotoxin A gen is investigated in 100 isolates and of 55 (%55) is found positive. 49 of 87 clinical samples (%56.32) are found positive while 6 of 13 (%46.15) natural samples are found positive.Adherence occurrence in Pseudomonas isolates: Bacteria adhered to 20 HE cells are counted under the microscope. Number of bacteria is found as 33 < 200, 52 between 200-300 and 19 >300 in clinical samples. In solid samples, the number of bacteria ,is found as 15<200 and 13 >200. In water samples the number of bacteria is found as 10<200 and 13 >200. Inhibition test for Pseudomonas bacteria, mannose galactose and N-acetyl glucosamine (NAGA) are used. The results of adherence tests are calculated as %. According to these results, mannose usage is found in 57-98 of clinical samples, 52-89 of solid samples and 55-87 of water samples. In galactose usage, it is found 48-84 in clinical samples, 51-82 in solid samples and water samples in 52-85. NAGA usage is found in 50-86 of clinical samples, 57-86 of solid samples and 54-92 of water samples.In conclusion it is indicated that occurrence of virulence factors has major role in the pathogenesis of bacteria.
