Abstract (EN)

ABSTRACT INVESTIGATION OF THE PROTECTIVE EFFECTS OF BENFOTIAMIN ON THE ALTERATIONS WHICH REVEALED WITH APPLICATION OF DOXORUBICIN IN THE RAT KIDNEY TISSUES Doxorubicin is an antracycline and shown to have nephrotoxic effects in different studies conducted in animal models and also in humans it is thought to have similar effects. In this study, we aimed to investigate the apoptotic changes in the kidney tissue with doxorubicin treatment and the anti-apoptotic effects of benfotiamine. In this study, 24 adult male Wistar Albino rats were used. Animals were divided into 4 groups as 6 animals in each group. During the 14-day experiment period, no application was executed to Group I. A single dose of doxorubicin in dose of 10 mg/kg (i.p) was administered to Group II; Group III received oral benfotiamine 70 mg / kg / day in addition to doxorubicin and to Group IV only 70 mg / kg / day benfotiamine was given orally. At the end of the experiment the rats were decapitated and kidney tissues were removed. Routine light microscopy follow-up were performed and tissues were embedded in paraffin blocks. Bax and caspas-3 immunohistochemical stainings were performed to tissue sections from the blocks. The findings examined and photographed. Light microscope examination of immunohistochemical stained specimens revealed marked increase of bax and caspase-3 immunoreactivity in doxorubicin group in comparison with control group and significant decrease was observed in doxorubicin + benfotiamine group. Tissue MDA levels were in accordance with the immunohistochemical findings. This study showed that doxorubicin induced apoptosis in kidney tissue and benfotiamine had anti-apoptotic and antioxidant effects against the effects of doxorubicin.
