Abstract (EN)

Early embryo selection criterias which used in the assisted reproductive techniques that is very important technics for embryo transfer. In our study we aimed to compare morphological criteria for embryo selection with genetic analys results. At the same time we aimed to show in the slow cleavage embryos day 5 ratio of fragmentation in blastomeres. In our study, in couples who could not have a baby in depite of there was not male factor it was applied in vitro fertilization on the methods assisted reproductive techniques. After the ovulation induction with ultrasound, when the oocyte came to desired size, they aspirated with the follicle liquids under anesthesia and these liquids was checked with microscope. At the same time same processes had done for selecting the active sperms of the sperm samples which had taken from the male. Oocyte cells extracted from surrounding granulosa cells by mechanical and chemical techniques and conjoined with sperm cells by microinjection methods. The embryos which had five or less blastomeres on the thirth day partipicated to our study. For taking one of the blastomeres ıt had shot with laser to the zones of embryos which were developing slowly. İt had applied FISH method with 5 probes to able to make aneuploidy analysis. After biopsy embryos were cultured until 5 th day. The embryos with the normal FISH results created the control group. Each normal and abnormal embryos after FISH fixed on the stage and TUNEL (Terminal dxynucleotidyl transferase mediated dUTP Nick End Labeling ) method was applied for showing the apoptosis in these slow developing embryos . As a result, it was investigated that if it was possible to select normal embryos with early selection parameters without requirement of PGD process. It was examinated that, the reason of slow development and anomalies of embryos whether the apoptosis or not.
