Abstract (EN)
Cancer is a hereditary illness characterized by disruption of normal cell division and growth. It spreads by mutations in cancer genes. Cervical cancer arises in the cervix cells (uterine lower portion). The aim of the study is to see how lipopolysaccharides (LPS) from Escherichia coli affect cervical cancer cells. The LPS was isolated from two E. coli strains, viz. BL21DE3 and DH5-α. The isolated LPS was quantified qualitatively. The gene expression profile and LPS action mechanism were examined in depth. E. coli BL21DE3 lipopolysaccharide was reported to be higher than E.Coli DH5-α. As a result, the LPS isolated from E. coli strain BL21DE3 was employed in the investigation. The LPS treatment raised Toll-like receptor 4 (TLR-4) expression, which boosted nuclear factor-κβ (NFκB), TIR Domain-Containing Adaptor-Inducing Interferon-β (TRIF), and Interferon Regulatory Factor – 3 (IRF3) expressions in SiHa and HeLa cervical cancer cell lines. The findings show that LPS not only promotes cancer development but also regulates inflammation and innate immunity. Immune evasion through TLR4-NFκB-LPS pathway should be researched in more depth. This pathway may lead to new medication development as a potential therapeutic target for cervical cancer.
