ABSTRACTMs. Sci. ThesisINVESTIGATION OF ANTI-ULCEROGEN AND ANTIOKSIDANT EFFECTOF Matricaria chamomilla L. ON ETHANOL-INDUCED ACUTE GASTRICMUCOSAL INJURY IN RAT MODELSEzgi YILMAZAfyonkarahisar Kocatepe UniversityGraduate School of Natural and Applied SciencesDepartment of ChemistrySupervisor: Assist. Prof. Dr. Mustafa CEMEKPeptic ulcer is a common disorder of the gastrointestinal system and millions of peoplesuffer from this disease in the world. The pathogenesis of peptic ulcer disease ismultifactorial, including chronically using non-steroid anti- inflammatory drugs, cigarette smoking, alcohol, and reactive oxygen species (ROS). ROS are generated bycells in some physiological and pathological circumstances. Any derangement betweenpro-oxidants and antioxidants, in which pro-oxidants prevail is known as oxidativestress. Insufficient antioxidant protection or excess production of ROS can result in thiscondition. Some studies have revealed that ROS and lipid peroxidation are implicatedin the pathogenesis of ethanol-induced gastric lesions and gastrointestinal damage, andthey attack and damage many biological molecules such as prostaglandins. Therefore, treatment with antioxidants and free radical scavengers can decrease ethanol-inducedgastric mucosal damage. In the present study, we have investigated possible anti-ulcerogenic and antioxidantactivities effect of extract obtained from aerial part of the Matricaria chamomilla L.(MCE) in ethanol-induced ulceric rats. Rats which were hungry remaining for 24 hoursof 5 groups were administered with 25, 50, 100, 200 and 400 mg/kg Matricariachamomilla L., one group was administered with 20 mg/kg famotidine, and groupcontrol was administered with same volume saline solution by gavage.Malondialdehyde (MDA) which is sign of oxidative stress and reduced glutathione(GSH) levels were measured in whole blood; ascorbic acid which is prove antioxidantproperty levels were measured in serum. The area of stomach tissue and gross lesionswere approximately calculated. The results were translated to the term of ?total ulcerarea/total gastric area? and these were expressed as an ulcer index. When blood samples controlled, it has been shown that MCE decreased MDA valuesand increased GSH levels. We found that vitamin C levels were increase as to controlgroup. In conclusion, MCE exhibited significant antiulgerogen and antioxidative effect inethanol-induced ulcers and augmented the antioxidant system in a positive manner.MCE may be a new alternative for clinical management of gastric ulcer diseases and/oran antioxidant against oxidative stress.2007, 106 pagesKeywords: Oksidative stress, Peptic ulcer, Ethanol, Antioxidant, Matricariachamomilla L.)
