Investigation of the effect of chronic pain induced by nevre ligation method on the oocyte count and maturity using nice

Abstract (EN)

Chronic pain, that is stated as an inescapable stress factor, significantly affects the life quality by the physical and mental functıon changes of patients. It is known that chronic pain interferes with hypothalamo-pituitary-adrenal axis and effects neuroendocrin systems.In this study, we aimed to investigate whether neurophatic pain as a chronic pain model affects the number and maturity of oocytes or not. The number and maturty of oocytes of the mice suffering neuropathic pain by using chronic constriction injury method was evaluated.4-8 week female Balb/c mice divided into naive control, sham operated and neurophaty groups were used in the experiments. Neurophaty was tested by using cold-plate test. Cold plate test was used to evaluate whether neuropathic pain was developed or not in the sciatic nerve ligated mice.After superovulation protocol was applied to all control, sham and NP groups, each group was divided two subgroups. First subgroup was housed in the same cage with male mouse following induction for breeding. The second subgroup were not housed with male mice. After induction, oocytes/zygote were isolated from ampullas of female mice which were housed with male mice and oocytes were isolated from ampullas of female mice which seperated kept apart with male mice. Classifying was done according to features of morphologically of Metaphase I (MI), Metaphase II (MII) and degenerate oocyte and zygote.According to results of experiments of chronic pain;- Cold plate latencies of mice, which become neuropathic by ligating sciatic nerve, was found meaningfully shorter than control and sham groups.- According to results of experiments of copulation rate, maturity and number of oocyte;- Total numbers of oocyst of neuropathic mice were lower than control and sham groups.- Values of oocyst maturation of neurophatic mice which were housed with male mice, are definitely lower than groups of control and sham with the same conditions.- Meaningful differences groups of control and sham as a result of being compared values of maturation according to housing together with male and single including theirselves of control, sham and NP groups.- As a result of being compared between control, sham and NP groups in the subgroups which were housed together with male mice, NP groups? copulation ratio was lower than sham and control groups.Consequently, lower level of CPL in the neurophatic mice indicated that threshold of allodynia was reduced, in other words, neurophatic pain was occured. Also, the differences between in the number of oocyte, oocyte maturity and copulation ratio in the neurophatic mice suggest that there may be an opposing effect of neuropathic pain on the oogenetic processes and fertilityKey Words: Mouse, chronic pain, maturation, oocyte, nerve ligation method
