Abstract (EN)
Lilium candidum is a perennial ornamental plant. Thanks to the active components in its content, it can be used in the treatment of many diseases. However, due to climate change and human factors, the plant, whose natural one is destroyed, is in danger of extinction. Due to this situation, it is aimed to protect the plant with different biotechnological methods in order to transfer it to future generations. One of these methods is cryopreservation. Cryopreservation is a technique that can provide longterm protection at low temperatures by using parts of the plant such as callus, meristem, pollen, and seeds. Thanks to this technique, many medicinal and endangered plants can be preserved in small areas. The aim of this study is to create the most suitable cryopreservation protocol for Lilium candidum plant and to ensure the continuity of the plant to future generations. In the study, firstly, callus and meristems were obtained from the stem bulbils of the plant by plant tissue culture technique. Then, the best time was tried to be determined by applying different PVS-2 application times (60, 75, 90, 105 and 120 minutes for callus, 30, 45, 60 and 90 minutes for meristem). Different nutrient media (MS, OM, WPM, LB, NB) were used with different concentrations of plant growth regulators (BA, NAA, GA3, Kinetin) to determine the most suitable one. As a result, the most suitable PVS-2 application time in calli was determined as 90 minutes and the most suitable nutrient medium was OM. It was determined that 60 and 90 minutes of PVS-2 application and MS nutrient medium were good in meristems.
