Abstract (EN)

Brucellosis is a zoonotic infection caused by Brucella bacteria, and direct or indirect contact with infected animals or their products is important in transmission. Vaccination is the primary measure tobe taken in preventing brucellosis. With the methods used in the production of Brucella vaccines, vaccine production is insufficient and requires a high level of labor. It has become a necessity toincrease industrial production by optimizing the mediums used in vaccine production. In recent years, stainless steel bioreactors have taken an important place in vaccine production and significantly affect the quality and production amount of the vaccine. Stainless steel bioreactors make significant contributions in providing the necessary environment (temperature, pH, oxygen, mixing speed) forbacterial growth in vaccine preparation. This study aimed to optimize the production of bacteria in liquid media for the Brucella S19 vaccine, which is currently being applied in cattle. Bacteria whose growth was completed on solid medium were passaged in three different liquid media in a sterile stainless steel bioreactor (1.medium: tryptose, yeast extract, glucose, NaH2PO4, Na2HPO4,2.medium: proteose peptone, yeast extract, glucose, NaH2PO4, Na2HPO4, 3.medium: Tryptone, yeast extract, glucose, NaH2PO4, Na2HPO4). The passaging process was continued in a 30% O2 environment, at 37o C, and at pH 6.8. The mixing speed was set between 300 and 420 rpm during passaging. During passaging, samples were taken at 0, 6,12, 24, 36, 48. hand viability counts were performed. Applications were performed triple orderfor each medium. The highest bacterial counts in the 1., 2.and 3. media at 48. h were determined as 10.2x1011, 7.1x109 and 6.0x1010, respectively. As a result of the comparison of three different media, we can say that the most ideal growth medium is tryptose, yeast extract, glucose, NaH2PO4, Na2HPO4.Our study is important in terms of contributing to vaccinations, which are the most important elements of preventive medicine
