Abstract (EN)

In this study, it is aimed to synthesize Fe3O4@TiO2 nanoparticles which combine photodynamic therapy and MRI contrast properties and to label them with technetium-99m (99mTc). For this purpose, multifunctional Fe3O4@TiO2 nanoparticles were synthesized. The surface modification of the synthesized structure was carried out by coating with tetraethyl orthosilicate and aminosilane. The functionalized Fe3O4@TiO2 nanoparticles were conjugated with lupulon derivatives (hexahydrolupulon and acetolupufenone) to increase anticancer potential and named as Fe3O4@TiO2-Si, Fe3O4@TiO2-Si-HLP, Fe3O4@TiO2-SiALP. Structural characterizations of Fe3O4@TiO2 nanoparticles and functionalized Fe3O4@TiO2 nanoparticles were determined by fourier transform infrared spectroscopy. Dynamic light scattering for the size distribution of nanoparticles, fluorescence spectroscopy for optical properties and fluorescence intensities, and scanning electron microscopy (SEM) were used for surface properties and size analysis. Diameter of the synthesized Fe3O4@TiO2 nanoparticles were observed approximately as 40 nm. The quality control studies of the 99mTc samples labeled was performed by using HPLRC (High Performance Liquid Radio Chromatography) and TLRC (Thin Layer Radio Chromatography) methods. Studies were conducted on healthy (RWPE-1 normal human prostate epithelial cell line) and cancer prostate cell lines (PC-3 human prostate cancer cell line) to determine in vitro biological behavior of the Fe3O4@TiO2-Si-HLP nanoparticles. As a result of the incorporation studies, it is seen that there is a time dependent change at the binding efficiencies Fe3O4@TiO2-Si-HLP to prostate cells over time. As a result, the percentage of binding of Fe3O4@TiO2-Si-HLP sample increased with time and reached approximately 11 % in 2 hours and no cell separation observed during the 4 hours of the experiment.
