Extraction of bioactive substances from peanut pine bark by pressurized hot water system

Abstract (EN)

In this study, the extraction of bioactive compounds from the bark of peanut pine trees was investigated by using a pressurized hot water extraction system (PWE). In the study, tree shells obtained from Çine region in Aydın city were used. Volatile matter, ash, lignin and cellulose were determination in peanut pine bark. The extraction conditions in which the maximum bioactive components in the pine bark would be obtained with the BSSE method were investigated by using the Central Composite Design (CCD) technique. In addition, the extraction processes of peanut pine bark with different methods described by other researchers were repeated in our laboratory under their optimum conditions. The results of the bioactive components (phenolic compound, flavonoid substance and antioxidant) under the defined optimum extraction conditions were compared with the BSSE. With the solution of the quadratic equation obtained as a result of the experimental design, the conditions for the highest target bioactive component have been revealed. As a result of the extraction with BSSE, the maximum total amount of phenolic substances in the extract per gram solid matter (182C temperature, 3,15 ml/min flow rate and 17,4 liquid/solid ratio) was 52,6 mg gallic acid found. Optimum conditions for the amount of flavonoid substance were determined at 99.5 0C temperature, 0.82 ml/min flow rate and 18,7 liquid/solid ratio and 124.3 mg Qe/g-km was obtained. The maximum of DPPH radical scavenging effect was determined at 99.5 C temperature, 2.5 ml/min flow rate and 18.7 W/W liquid/solid ratio and it was obtained as 472.1 mmol trawl/g-km. For maximum ABTS radical scavenging effect determination, optimum conditions were determined at 99.5 0C temperature, 3.83 ml/min flow rate and 18.7 W/W liquid/solid ratio and 542.3 mmol trawl/g-km was obtained.
