Determination of the compositions of Ganoderma lucidum fractions extracted by conventional and supercritical extraction methods

In these study, Identification of bioactive component from the fruiting body of two different local Ganoderma lucidum strains (Balcali and Alata) were obtain with a sequential extraction procedure. As a preliminary clean up step, apolar components like fatty acids were extracted via supercritical CO2 atmosphere at 50ºC and 3300 psi pressure. Fatty acid composition of extracts were identified from fatty acid methyl ester standarts with GC. At the next stage extraction of triterpenoids from these local G. lucidum strains were carried out with ethanol under reflux. The components of these groups were fractionated with a semi-preperative HPLC system. Chemical structure identification of these fractions were carried out with HPLC/MS-MS (tandem ms) and 1H NMR measurements.In the last step, water-soluble polysaccharides and polysaccharide?protein complexes were extracted from fruiting bodies with hot water. Molecular weight characterizations of these strains were determined with both (HPSEC-MALLS) and GFC system. Aggregation of polysaccharide molecules in extract was also revealed by HPSEC-MALLS analysis. The effects of heating and sonication on the solubility of aggregates were also investigated. Protein%, Uronic acid% and monosaccharide content analysis were also applied on these extract.
