Abstract (EN)

In this study, we aimed that determine antioxidant, antimicrobial and DNA protective activity of Helvella leucomelaena and Sarcosphera crassa fungus species with ethanol extract. To determine antioxidant activity Rel Assay Diagnostics kits were used (TAS, TOS) and DPPH method. For the test of antimicrobial activity 8 different bacteria species (Staphylococcus aureus ATCC 25923, Pseudomonas aeruginosa ATCC 27853, Klebsiella pneumoniae ATCC 700603, Staphylococcus aureus ATCC 29213, Escherichia coli ATCC 25322, Escherichia coli ATCC 35128, Escherichia coli ATCC 10799, Escherichia coli ATCC 8739) were used and studied with minimum inhibition method (MIC). DNA protect activity was determined with agarose gel electrophoresis. According to the results, not only DPPH but also Rel Assay Diagnostics kits didn't clean so much free radical in the extract of Helvella leucomelaena and Sarcosphera crassa. When the antimicrobial test results were evaluated, it was determined that the extract of S. crassa more effective than the extract of H. leucomelaena. Also it was found that the applied concentrations have no protective activity against to pBR322 plasmid in the test of DNA protective activity. Consequently, it was determined that the extracts which obtained both fungi species have antioxidant and antimicrobial activity but no have protective effect on DNA. Keywords: Helvella leucomelaena, Sarcosphera crassa, Antioxidant, Antimicrobial, TAS, TOS, DPPH, DNA protective activitiy.
