ABSTRACT INVESTIGATION OF THE MUTATIONS ON S GENE REGION OF HEPATITIS B VIRUS' GENOTYPE D SEROTYPE AND RESEARCH OF THIS MUTATIONS' EFFECTS ON THE TRANSCRIPTS AMONG THE DIFFERENT PATIENT GROUPS BARSGAN, Arzu M. Sc. In Biology Supervisor: Assoc. Prof. Dr. Mehmet ÖZASLAN September 2004, 124 pages Hepatitis B virus is an agent causing the Hepatitis B that important public health problem. HBV causes liver cirrhosis and hepatocellular carcinoma. In this study 40 HBV-DNA positive patients were determined from among 132 HBsAg positive patients by PCR. All varients were designated Genotype D and subtipe ayw3 using the semi-nested PCR and DNA sequencig thecniques datums. This patients consisting of different groups. According to these, mutations datums were investigated in the patient groups and the rates were determined. In addition effects of these mutations on the transcripts were researched. For these studies S gene region that a stable region against mutations and responsible expression of surface antigens, was worked. In chronic patients and family members' mutation rates were designated as maximum among other groups. Aminoacid mutations' maximum rates were occured in same groups. The one of the most important consequences is 125.(M- >T) and 127.(T- >P) aminoacid mutations that present all of the varients. And these mutations were founded on the first loop of a-determinant.
