Abstract (EN)

Breast cancer is the most common type of cancer in women and ranks second in the world among the causes of female death. In order to find a solution to breast cancer, different studies conducted for treatment and the effects of different drugs and substances on this disease investigated intensively. In this study, it was aimed to investigate the antiprolipheratif, apoptotic, angiogenic and antioxidant effects of boric acid use in breast cancer cell line (MCF-7). Chemotherapeutic agents damage neoplastic cells, while some normal cells are affected, causing various side effects. Boric acid has been shown to control the proliferation of certain types of cancer cells. Boric acid is an inhibitor of peptides, proteases, proteasomes, arginase, nitric oxide synthase and transpeptidases. Boric acid has a strong antioxidant effect, especially in reducing the side effects of drugs used in cancer treatment and reactive oxygen species (ROS) is important to reduce the level. In this study, different concentrations of boric acid were applied on the MCF-7 cell line to determine the mean latel dose (LD50) values of the boric acid and the antiproliferative effect was determined using the cytotoxic activity of the cell count (CCK-8) ELISA kit at iv 24, 48 and 72 hours. The most significant changes in the results were seen at 48 hours. For this reason, other analyzes were made over a 48-hour incubation period. After the concentrations determined for the determination of biochemical parameters were applied to the cells, cell lysates were obtained. Vascular endothelial growth factor (VEG-F) elisa kit was used to determine angiogenic effects with the samples obtained. In order to determine apoptotic effects, Poly (ADP-ribose) polymerase (PARP) elisa kit was used. TAS and TOS elisa kits were used to determine total antioxidant and oxidant levels. As a result, in this study, it was observed that the concentration and time-dependent boric acid decreased proliferation in the MCF-7 breast cancer cell line. At the same time, according to the PARP results we obtained, it was determined that the boric acid concentrations showed a significant difference compared to the control group. No significant difference was observed between boric acid concentrations and Cis Platin groups. The increase in both groups compared to the control group suggests that boric acid leads the cell to necrosis. When looking at the results of VEG-F performed to explain the relationship of boric acid with angiogenesis, significant differences were observed with both control group and Cis Platinum. In the analysis, more effective results were obtained from Cis Platin in the MCF-7 breast cancer cell line in boric acid groups, suggesting that boric acid may have an antiangiogenic effect. As a result of TAS-TOS analysis, the anthoxidant capacity and oxidant capacity increased due to concentration. This suggests that the cell undergoes oxidative stress and in turn increases antioxidant levels in order to balance. From this point of view, we believe that the dose-dependent changes of biochemical analyzes in MCF-7 cells will contribute to the literature. Keywords : MCF-7,Boric Acid, PARP, VEGF
