Abstract (EN)
Prostate cancer is one of the most deadly cancers in the worldwide. İt is has been found that, a variety of ion channels affect mechanisms such as proliferation, angiogenesis, invasion, cell differentiation, cell volume growth, tumor growth, metastasis and apoptosis in cancer cells. It is considered as a potential targets for cancer therapy due to the ion current resulting from ion channel activity is a effective molecules in the mentioned basic mechanisms. In consequence, we are aimed to investigate the ion channel expression levels in prostate cancer tissues by comparison normal prostate tissues. A total of 165 cases that are formalin fixed paraffin embedded 105 patients that was diagnosed prostate adenocarcinoma [n=105], 30 benign prostatic hyperplasia [BPH] patient tissues [n=30] and 30 being normal prostate tissue [n=30] evaluated in our research. Quantitative Real Time-PCR method was used for mRNA expression analysis after RNA isolation. TRPC1, TRPC3, TRPC4, TRPC5, TRPC6, TRPC7, TRPV2, TRPV3, TRPV4, TRPV5, TRPA1, Cav3.1, Cav3.2, Cav3.3, Kv1.3, Kv1.4, Kv1.6, Kv4.3 , Kv11.1, KCa1.1, KCa3.1, TASK-3 [KCNK9] and Clcn-3 ion channels expressions were analyzed in all samples. The quantitative RT-PCR results showed that there was no significant change in the KCNA3 gene expression in the other groups except for a significant decrease in gleason scores 6 and 7. When KCNA6, KCNK9, KCNMA1, and KCNN4 gene mRNA expressions were compared to controls, it was found that this ion channels has a significant decreases in all cancer tissues except for BPH in KCNA6. While TRPC3 mRNA expression compared with control's has a significant increase in only gleason score 10, it was found that important increases in TRPC3 and TRPC4 mRNA expressions in all groups. It was established that meaningful decreases in the mRNA expressions of TRPC5, TRPA1, TRPV3 and TRPV4. As compared with control, while CIC-3 gene expression has a significant increase in gleason score 7, there was a significant decrease in gleason score 10. Prostate cancer groups compared with controls were not found meaningful difference in TRPC7, CACNA1G and KCNA4 mRNA expressions. Alterations in ion channel mRNA expressions may lead to prostate carcinogenesis by contributing to many transformation processes such as increased proliferation, apoptosis resistance, metastasis, and invasion. The data we obtain are preliminary findings and the contribution of each channel to the cell transformation process needs to be examined and verified with further researches.
