Investigation of ndm enzymes in Escherichia Coli in women and infant children with urinary tract infection in Diyala / Iraq

Abstract (EN)

This study was aimed to phenotypic and genotypic detection of NDM genes in multidrug resistance Escherichia coli isolated from women and infant children with urinary tract infection. VITEK-2 uesd for the antibiotics sensitivity. PCR were used for molecular detection of bla NDM1, bla NDM2, bla TEM, bla VIM, bla IMP genes. On the other hand Phenotypic detection were used to identify some virulence factors like, metallo, β-Lactmase and biofilm formation. A 200 clinical samples were collected from hospitals in diala-iraq, the ages were 2.5 to 59 years. MacConkey agar,blood agar and EMB agar cultures were used for holding the samples to the bacteria MDR test, molecular tests. As a results Only 49 urine samples were positive for E. coli, (27.08%) infants and (72.92%) females. The isolates showed high levels of resistance to ceftazidime (100%) and ampicillin (100 %), followed by cefepime (97.96%), cefotaxime (97.96%), and Amoxicillin-clavulanic acid (95.91%), while the lowest level was for imipenem (20.90%). Meanwhile the resistance genes in isolates was bla-TEM gene with highest frequency at (83.33%), bla-SHV (39.58%), bla-VIM (25%), NDM-1 (37.50%), On the other hand, none of the isolates tested positive for the bla-TMP gene. All of the E. coli isolates have the capacity to build biofilm, around (91.67%) of the total isolates. For phonotypically perspective out of 49 E. coli isolates (31.25%) were positive for MBL, in contrast (68.75) were negative for MBL, out of 16 MBL positive isolates only 5 isolates were harbor bla-NDM-1 gene, out of 15 of MBL positive isolates only 11 isolates were harbor bla-TEM gene, while out of 16 MBL positive isolates only 6 isolates were harbor bla-SHV gene, out of 16 MBL positive isolates only 6 isolates were harbor bla-VIM gene.
