Abstract (EN)

Integument which covers body surface of insects is an ectodermic originated and consist of three parts; basal membrane, epidermis, and cuticula. Epidermis has an important characteristic properties in the larval life period, in pupal phase and in metamorphic changes. Integument of Lepidoptera is soft and elastic in larval period, solid and protective in pupal phase, covered with lamina in adult period. All these properties are produced by same integument cells. When we think that these changes are taking place by the ecdysone receptor complex, the role of the juvenile hormone and the management of it get a vital importance. In Insects that have an whole metamorphose like silk warms, the metamorphose processes are controlled by two important secretions; 20-hydroxyecdysone which has steroid character and juvenile hormone. 20-hydroxyecdysone induces morphological and biochemical changes in which occur in larval pupa and pupal-adult transformation and molting. In case of Juvenile hormone, it makes to have same larval characteristics working as ecdysone antagonistic. In this thesis, the morphological characteristics of epidermis are presented using histological cross-sections in Bombyx mori which in the last larval stage, during larval-pupal metamorphosis and pupal phase. Fenoxycarb (ethyl[2-(4-phenoxy-phenoxy)-ethyl]carbamate) which is analogue of juvenile hormone is performed on the third days of fifth larval phase. The histological effects of Fenoxycarb on epidermis is established in larval-pupal development stage. Also, density and localization of ecdysone receptor B1 which is one of the ecdysone receptor isoforms is determined by immunohistochemical methods. Nevertheless, the total protein profile of the epidermis is revealed by SDS-PAGE method. As a result, the application group has not shown spinning behavior, and molting has not took place by extending larval phase. Besides, absence of localization of ecdysone receptor B1 is presented or may have a tiny amount that does not make sense.
