Abstract (EN)
In the first part of our study, electrochemical biosensor was developed with ionic liquid modified single use pencil graphite electrodes (PGE) for detection of miRNA. Surface characterization of ionic liquid modified by scanning electron microscopy (SEM), electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV) techniques was performed. The selectivity of the biosensor was investigated against to other miRNA sequences. In addition, the determination of miRNA-34a in buffer medium and testing of the selectivity of the biosensor, as well as the determination of miRNA-34a in the artificial serum medium was performed and the selectivity of the biosensor was tested. In addition, the applicability of biosensor developed for voltammetric miRNA analysis in total RNA samples isolated from HUH-7 cell line was investigated. In the second part of our study, electrochemical biosensor was developed by ionic liquid modified PGE for detection of globulin. Globulin, type of globular protein, was determined by DPV technique. The surface characterization of ionic liquid modified electrodes by electron microscopy (SEM), and electrochemical characterization of ionic liquid modified electrodes by electrochemical impedance spectroscopy (EIS), cyclic voltammetry (CV) techniques were performed. The selectivity of the developed biosensor was also tested in the presence of similar biomolecules such as glutathione (GSH), glucose, L-Cysteine (L-Cys), L-Methionine (L-Met), L-Tryptophan (L-Trp). In our study, electrochemical biosensors developed for the detection of miRNA and protein by using ionic liquid modified single use pencil graphite electrodes offer many advantages in terms of high sensitivity, ease of use, short analysis duration, non-toxic experimental steps and low cost. The obtained results show that the combination of biomolecules on conductive materials such as, ionic liquids effectively increases the performance of the electrochemical biosensor. Therefore, there is an increase at the potential applications of ionic liquids for biomedical and bioengineering studies in the development of advanced biosensor technologies.
