Production of amphidiploid with effect of colchicine in natural hybrid of tall fescue (Festuca arundinacea schreb) x perennial ryegrass (Lolium perenne L.)

THESIS) FATMA ÜNAL GAZI ÜNIYEESITI INSTITOT?E OF SCIENCE, AND TECHNOLOGY JANfAET 1988 In this study it was investigated to obtain amphidip- loid plant with effect of colchicine in natural hybrid of tall fescue (Festuca arundinacea Schreb.) x perennial ryegrass (Lolium nerenne L»), The chromosome numbers hare been found 2n-42 in tall fescue, 2n=14 in perennial ryegrass and 2n=28 in hybrid which was obtained from these two plants* The sterile plant which was obtained from natural hyb ridization has been treated with colchicine solution for 24- hours at 18-20°C. Amphidiploid clones with 2n=56 chroEOsoEes hare been obtained at the ratio of 20-30 %» Triangular incision has been made at the base of the clones and the same applications have been repeated in 0.4 # colchicine solution which was dissolved in 2 # dimethyl-sulphoxide (MSO). Finally, aaphidiploid clones with 2n=56 chromosomes, have been obtained at the ratio of 80 #. IVDuring the preparation of the elides, the pretreat- ment of root tips hare been treated with saturated oC-mono- bromonaphthalene solution at 4°G for 16-17 hours. The fixation and hydrolysis have been made in glacial acetic acid at the j?oom temperature for half an hour and in 1 H HC1 at 60°C for 10-12 minutes, respectively. The slides have been made according to squash technique. After the treatment of alcohol steam, they were mounted in Euparal, The measurements of chromosomes length are given below: 4,826-2,472 jK in tall fescue, 4,761-2.838/* in perennial ryegrass, 5« 669-2. 561 JX in natural hybrid, 5.196-2.772/1 in amphidiploid. The karyotype analysis and idiograms of parents, hybrid and amphidiploid plants were made.
