Abstract (EN)

The first discovered psychopharmacological agent in the light of historical written sources is alcohol (ethanol). This is why it is one of the most commonly used addictive substances. Alcohol consumption can lead to many illnesses and the most noticeable of these diseases is toxic effects on the liver. It is known that in the past and nowadays many plants and also their seeds have been used in the treatment of different diseases. In this thesis, nanoemulsions (A1 and A2 formulations, with and without GSH, respectively) with different contents were prepared using Nigella sativa seed oils obtained by supercritical carbon dioxide extraction. After the various contents and characterization tests of the nanoemulsions were made, a number of in vitro cell culture experiments were studied to prevent and eliminate the alcohol-damages on THLE-2 cell lines, which is non-tumorigenic and represents phenotypic characteristics of normal adult liver epithelial cells. As a result of this thesis, especially for hepatoprotective activity promising results were obtained in the field of addiction, which was the aim. In the experimental stage, firstly, content analyzes and characterization studies of prepared nanoemulsions were carried out. Then doses to be used in in vitro cell culture experiments were determined (0.5 mg/mL).After that, wound healing, proliferation, antioxidant activity and fluorescence imaging studies of nanoemulsions were carried out in cell culture in vitro conditions. While investigating the hepatoprotective activity of nanoemulsions, the main goal of the study, THLE-2 cells were exposed to ethanol at different time intervals and concentrations to determine the ethanol concentration to be used throughout the study (400 mM). In order to investigate the protective effects, pre-treated cells with 0,5 mg/mL nanoemulsions for 2 hours were treated with 400mM EtOH for 48 and 72 hours and cell viability test was performed. The hepatoprotective effects of the A1 and A2 formulations on the THLE-2 cell line, which was used as a model, were proved in the light of the results.
