Abstract (EN)

In this study, it is aimed to investigate the antimicrobial, antioxidant, cytotoxic activities and DNA interactions of Klasea bornmuelleri (Azn.) methanol, ethanol and water extracts. To this end, the extracts were analysed for antimicrobial activity by agar well diffusion and minimum inhibition concentration (MIC). It was observed that methanol and water extracts had mild antimicrobial activity against some tested bacteria. The methanol extract was observed to be effective against E.coli ATCC 25922, E.coli ATCC 35218 and S.aureus ATCC 25923 bacteria. In addition, it was noted that the water extract was also effective against K. pneumoniae ATCC 25955. The methanol extract showed activity against the maximum number of microorganisms and created the largest zone of inhibition among the extracts studied was found to have the most effective antimicrobial activity. The extracts were analysed for antioxidant activity by 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging and phenolic content detection with Folin-Ciocalteu reactive. According to the results, the ethanol extract (IC50: 50,5±0,5 µg/ml) had DPPH radical scavenging activity and phenolic content higher than methanol (IC50: 83,5±0,5 µg/ml) and water (IC50: 91,7±0,5 µg/ml) extracts. The interaction between the extracts and pBR322 plasmid DNA were analyzed by agarose gel electrophoresis. It was found that the methanol and ethanol extracts, depending on the concentration of compounds and the duration of incubation, showed effects such as DNA destruction and cleavage. Cytotoxic effects of the extracts on HeLa (Cervix Cancer) and MCF-7 (Breast Cancer) cells were detected by 3-(4,5-dimetilthiazol- 2-yl)-2, 5-difeniltetrazolium bromid (MTT) method and also by propidium iodide (PI), 4',6-diamidino-2- fenylindole (DAPI) and 3,3-dihexyloxacarbocyanine iodide (DiOC6) fluorescence dyes. The ethanol and water extracts at 0,001-100 ?g/ml concentration range were effective on HeLa (Ethanol IC50: 9,84±0,05 ?g/ml, Water IC50: 0,1±0,005 ?g/ml) and MCF-7 (Ethanol IC50: >100 ?g/ml, Water IC50: 11,04±0,05 ?g/ml) in 24 hours. In fluorescence dyes results, it was found that ethanol extract had an apoptotic effect on HeLa cell line. As a consequence, it was detected that HeLa was the most sensitive cell line and was more sensitive against ethanol extract than MCF-7.
