Abstract (EN)

Lipases (triacylglycerol acyl hydrolases) in the E.C.3.1.1.3 class in the classification made by the Enzyme Committee are serine hydrolases, which are part of the hydrolase family that has the task of breaking down triglycerides into diglycerides, monoglycerides, fatty acids and glycerol. In this study, it was aimed to purify and characterize the lipase enzyme from sheep pancreas. After degreasing sheep pancreas, 16.2 fold purified lipase was obtained by ammonium sulfate precipitation and gel filtration chromatography methods. After each purification process, protein determination was made using the Lowry method and lipase activity was determined using the titration method with NaOH. The purified lipase enzyme showed the highest activity at pH 8.0 and 40°C. Activity measurements were carried out for one week for the storage stability of the purified lipase. To determine the Km and Vmax values of sheep pancreatic lipase, triolein was used as substrate and calculated as 76,86 mM and 22,98 U/min.mg.enzyme, respectively.
