Interleukin-6 and interleukin-10 polymorphisms in chronic lymphoid leukemia

Abstract (EN)

Chronic lymphoid leukemia (CLL) is a leukemia characterized by an abnormal proliferation of mature small monoclonal B-lymphocytes in peripheral blood, bone marrow, and lymphoid tissue. Cytokines are molecules that play an important role in regulation of the immune system and inflammatory events in the organism. A significant proportion of the cytokines secreted from the immune system are interleukins (IL) and their main tasks are to stimulate the immune system cells. Therefore, the genotypic effects of these interleukins; IL-6 and IL-10 in the immune system in CLL were investigated in this study. For this purpose, 100 patients with the diagnosis of CLL and 70 healthy subjects were included in this study. DNA was isolated from the blood samples obtained from the CLL patients and control groups. Polymorphisms in the IL-10 (1082 A/G and 819 C/T) and IL-6 (174 G/C) genes were studied by RT-PCR. Genotype and allele frequencies were calculated by directly. The statistical significance of genotypic distributions between CLL patients and control groups was evaluated by chi-square test. IL-10 1082 A/G region, 45 wild-type AA, 40 AG and 15mutant-type GG genotypes were detected in 100 CLL patients. Genotypic distribution of 819 C/T regions of IL-10 was detected in CC in 37 patients, CT in 50 patients and TT genotype in 13 patients. IL-6 174 G/C region was detected in GG in 62 patients, GC in 30 patients and CC genotypes in 8 patients. There was no statistically significant difference between CLL patients and control groups of IL-10 1082 A/G and 819 C/T and IL-6 174 G/C regions (p> 0.05). Allele frequency of IL-10 1082 region for the patient group was determined as 0.65 (for A) and 0.35 (for G), while these values were determined to be 0.71 and 0.29 for the control group. The frequency in the patient group of 819 was determined as 0.62 and 0.38 for C and T, while the values were found to be 0.61 and 0.39 for the same bases in the control group. In the case of IL-6 174 group, frequencies of C and G were found as 0.77/0.23 and 0.79/0.21 in the patient and control groups, respectively. Considering the number of patients discussed in this study, genotype frequencies of IL-10 and IL-6 are thought to be not statistically related to CLL patients. Mutant alleles of all interleukin SNPs were found to be higher in the patient group than the control group. Therefore, in future studies, by increasing the number of samples, the possible relationship between SNPs of these interleukins and CLL can be determined.
