Abstract (EN)

Lactoferrin (Lf) is a glycoprotein which is found in milk and has significant biological activities. In this thesis, improving of a new generation cross linked anti-lactoferrin (anti-Lf) antibody cryogel column and using it in affinity chromatography have intended for the purification of Lf which is found in body fluids, a defense protein containing iron and inhibits growth of tumor from cow milk. For this purpose, cross-linked cryogel column which contains anti-Lf in its pores has synthesized. P(Anti-Lf-co-HEMA) cryogel has used as column filling (extender) material and Lf separation has provided by Fast Protein Liquid Chromatography (FPLC). The purity of Lf obtained from cow milk has investigated by SDS-PAGE. The swelling test, FT-IR, SEM and BET analyses have applied for the characterization of the column. As a result; the columns which have high selectivity for Lf and are reusable have improved for the first time using photosensitive conjugation method (ANADOLUCA) which has WIPO and USPTO patented by R. Say. These developed columns will be an alternative for the purification of Lf which is an important agent for cancer, vaccine and imaging technologies.
