Abstract (EN)

This study was carried out between 2018-2019 in Malatya Turgut Özal University Faculty of Agriculture, Department of Horticulture, Plant Biotechnology Laboratory. The objectives of this study are to determine the resistance of Banazkara and Köhnü grape varieties, which have an important place for the local economy of Malatya, to the biotic and abiotic stress conditions, to add new features to their superior qualities, to support the breeding studies to be carried out in the next period to obtain virus-free plant production, to shorten and improve the breeding processes and to provide the basis for biotechnological studies for their improvement and to maintain them as genetic resources if needed. In the study, the culture was initiated by applying surface sterilization to micro cuttings taken from the shoots and transferred to MS medium containing 30 g/l sucrose and 1 mg/l BAP. The effects of different mediums, MS medium strength, different sugar types, BAP strengths, and different doses of NAA/IBA added to BAP-supported medium were examined using the 0.5-1 cm shoot-tips obtained. The effects of auxin group plant growth regulators were investigated to determine the rooting capabilities of the shoots. At the end of the experiments, the number of shoots, shoot length, the number of nodes, root number, and root length parameters were examined. According to the observations made and the results obtained, for proliferation in Banazkara variety, MS medium + 30 mg/l sucrose + 0.75 mg/l BAP and / or (0.6 mg/l BAP + 0.1 mg/l NAA) for rooting medium on the other hand, 1mg/l NAA has been determined as best. For the proliferation of Köhnü variety, MS medium + 30 mg /l sucrose + 1 mg /l BAP and/or (0.6 mg/BAP + 0.3 mg/IBA); and for rooting, 1 mg / l NAA determined as the most suitable.
