Investigation of metabolomic biomarkers for detection of benign and malign ovarian tumors and platinum-sensitive and platinum-resistant ovarian cancer

Abstract (EN)

In this study, it is aimed to determine ovarian cancer patients with platinum resistance for alternative treatment protocols by using metabolomics methodologies. Moreover, early diagnosis of ovarian cancer and differentiation of cancer patients from benign tumors were also aimed to determine by metabolomic biomarkers. The defined biomarkers were identified and validated. Metabolomic pathways related to group separations were also specified. Urine and serum samples of platinum resistant ovarian cancer, platinum sensitive ovarian cancer, benign tumor and healthy controls were analyzed using GC-MS. The biomarkers were verified after univariate, multivariate and ROC analysis. Finally, metabolomic pathway analyzes were performed. R2Y, Q2, AUC, sensitivity and specificity values of platinum resistant and sensitive ovarian cancer patients' serum and urine samples were 0.85, 0.545, 0.844, 91.30%, 81.08 and 0.570, 0.206, 0.743, 77.78%, 74.28%, respectively. Metabolomic analyses performed in urine and serum samples of patients with malignant and benign tumors yielded results of R2Y, Q2, AUC, sensitivity and specificity values as 0.719, 0.482, 0.887, 83.50%, 92.53% and 0.488, 0.091, 0.843, 79.54%, 79.00%, respectively. According to urine and serum samples of malignant patients and healthy controls, R2Y, Q2, AUC, sensitivity and specificity values were found as 0.870, 0.773, 0.987, 91.75%, 97.18% and 0.809, 0.748, 0.990, 93.18%, 96.77%, respectively.
