Abstract (EN)

In order to determine the Barley yellow dwarf virus (BYDV-PAV, BYDV-MAV, BYDV-SGV, BYDV-RMV and CYDV-RPV) in Mardin province, survey studies were conducted in wheat production areas in 2017. In this context, four hundred samples were collected in survey areas and tested by Multiplex RT-PCR and RT-PCR. According to the test results it was found that BYDV-PAV in 10 samples (2.5 %), BYDV-SGV in 7 samples (1.75 %), CYDV-RPV in 2 had (0.5 %), and BYDV-RMV in 1 (0.25 %). Seven of the positive isolates were found to be infected by BYDV-PAV + BYDV-SGV (1.75 %) and 1 of them had BYDV-PAV + CYDV-RPV (0.25 %) mixed infection. The overall infection rate of wheat viruses in Mardin province was found to be 3.0 %. None of the 400 wheat leaf samples tested had BYDV-MAV detected. In this study, BYDV-PAV, BYDV-SGV, BYDV-RMV and CYDV-RPV viruses were detected for the first time in Mardin province. A random isolate was selected from each detected virus to characterize the coat protein gene regions, which of complete for BYDV-PAV (Genbank accession no.MK732034) and CYDV-RPV (Genbank accession no.MK732035) viruses and partial for BYDV-SGV (Genbank accession no. MK940529) and BYDV-RMV (Genbank accession no. MK955886). The coat protein gene region of selected izolates were cloned and sequenced with the universal primers.
