Abstract (EN)
Myrtle (Myrtus communis L.) is one of the important aromatic and medicinal species from the Myrtaceae family. Myrtle is a widely used spice that has many applications in the perfumery, food and pharmaceutical industries. The RNA-Seq technique is a recently developed advanced transcriptome analysis technique. Next generation sequencing technologies are often used to detect and develop SSR markers in recent years. In this thesis, it is aimed to develop SSR markers using next generation sequencing technologies. For this purpose, RNA sequencing studies were carried out using Illumina HiSeq 2000 Sequencing device on two myrtle genotypes, one with black fruits and one with white fruits. Over 111.000 transcripts were obtained in the samples. The total length of the transcripts ranged from 79.141.165 to 93.880.054. The number of unigenes detected in the samples was determined as 68.476-79.802. As a result of RNA-Seq analysis, repeating regions on the genome were determined by de novo analyzes performed on two different myrtle genotypes. As a result of the findings, the highest repeat motif was found to be 12.885 di-nucleotides. In addition, the number of different repeating motifs ranging from 12-11776 were revealed. Suitable SSR primers were designed to amplify these identified repeat motifs. As a result of this study, over 10.000 SSR primers were developed.
