Investigation of loss of heterozygosity on chromosome 4q22-35 region in patients with bladder cancer

Abstract (EN)

Cancer is a disease that occurs by losses of function of the genes controlling or accelerating the cell division or by their being highly active. Deletion of tumor supressor genes or decreasing their expressions can be shown among the genetic or epigenetic reasons of cancer. Loss of heterozygosity (LOH) analysis is an effective method used in determining the candidate tumor supressor gene areas peculiar to the cancer type. Urinary bladder cancer is a disease affecting lots of people in our country and all over the world and causes to death of thousands each year. It has drawn attention that in the genome screening studies of urinary bladder cancer cases done previously; there is a surplus of the mycro losses on the areas close to telomere on the long chol of the fourth chromosome. However, a detailed loss mapping of these areas couldn?t be done. In this study, chromosome 4q22-35 area was analysed by using 13 mcyrosatellite markers in 25 patients with urinary bladder cancer. In the analyses three loss areas have attracted the attentions. It was identified that the first area is D4S1564 marker and its around, second area is D4S1644 marker and its around and the third one is D4S2979 marker and its around. As a result, in urinary bladder cancer cases for chromosome 4q22-35 area, CASP6, DDK2, SAP30, CASP3, SMARCA5 and ING2 genes are specified as candidate tumor supressor genes.
