Abstract (EN)
ABSTRACT DETERMINATION OF THE EFFECTIVE RADIATION DOSE FOR MUTATION BREEDING IN PURPLE CARROT AND POSSIBLE VARIATIONS FORMED YARAR, Gülistan M.Sc. Thesis. Department of Agricultural Biotechnology Supervisor: Assoc. Prof. Dr. Mehtap YILDIZ August 2019, 68 pages The effective gamma mutation dose in purple carrot was aimed to determine in this thesis which was conducted. For this purpose, different gamma irradiation doses [0, 50, 100, 200, 300, 400, 500, and 600] were applied to the seeds of the local Hatay purple carrot population. Purple carrot seeds were planted in pots and survival rates were revealed by regression analysis. The effective dose of mutagen was shown as 387.5 Gy. Root length, shoulder diameter and length of root's green parts of control and mutant plants which were exposed to different doses were measured and it was found those characters were affected adversely in mutant plants. SRAP and ISSR markers were used to determine the polymorphism among the control and mutant plant groups. A total of 100 bands were obtained using 4 ISSR primers and 9 SRAP primer combinations, of which 84 (84%) were polymorphic. Using SRAP markers, the highest number of bands (13) was obtained from Me2 X Em16 primer combination at 100 Gy and 400 Gy doses, while the lowest number of bands (2) was obtained from Me1 X Em3 combination in the control group. On the other hand, when ISSR markers were used, the highest number of bands (10) was obtained from ISSR3 primer at 50 Gy and 100 Gy doses, while the lowest number of bands (4) was obtained at 600 Gy dose from Sola11 primer. The dendrogram created based on the similarity matrix of JACCARD consisted of 3 main groups; a) control group, b) a group of mutants exposed to 600 Gy dose, c) The third group included two subgroups. The mutant plants which were exposed to 50 Gy, 100 Gy and 200 Gy were collected in one subgroup, 300 Gy, 400 Gy and 500 Gy applied mutant plants were collected in another subgroup.
