The association of pathophsiological changes in menopause and oxidative stress parameters with serum sVCAM,sE-selectin,leptin levels in postmenopausal women taking ERT(estrogen replacement treatment) and HRT(hormone replacament treatment)

olarak K grubunda saptanmış olup, post-menopozal druplar ilekıyaslandığında fark istatistiksel açıdan anlamlıdır. NO düzeylerinin ise, Kgrubunda en düşük düzeyde olduğu, diğer gruplarda ise yüksek olduğubulunmuştur. Ortalama MPO aktivitesi ise her dört grupta yaklaşık aynıdeğerde saptanmıştır. Gruplarda, kardiyovasküler belirleyiciler olansVCAM-1 ve sE-selektin düzeyleri arasında fark saptanamamış olup, budurum olgularımızın hiçbirinin koroner bir yetmezliği olmaması ve lipidprofillerinin düzgün olması ile açıklanabilmekte olup, inflamatuar birbelirteç olan MPO bulguları da bu konuda çalışmamızı desteklemektedir.Obezitenin bir belirleyicisi olan leptin düzeylerinin, aynı zamanda östrojenve BMI (kg/m2) ile ilişkisini değerlendirdiğimizde, ERT ve HRT grubundaortalama leptin düzeylerinin FM grubu ile kıyaslandığında daha yüksekolduğu ve östrojenin leptin düzeylerini arttırdığı saptanmıştır. Yine post-menopozal obez grupta (FM) leptin düzeylerinin obez olmayan FM grubuile kıyaslandığında daha yüksek olduğu saptanmıştır. Sonuç olarak,çalışmamızda menopozun oksidatif sistemi bozduğu, lipidlerinperoksidasyonuna neden olduğu, GSH metabolizmasını etkili olduğusaptanmış olup, ölçtüğümüz parametreler arasında ERT ve HRT alangruplar arasında anlamlı bir fark gözlenmemiştir.7. SUMMARY (İNGİLİZCE ÖZET)Estradiol (E2) is a molecule which has antioxidative properties. Therole of prOgestins in antioxidant defense is stil unknown. We evaluatedthe influence of E2 and E2 + progestin on erytrocyte antioxidative enzymesystem activity and adhesion molecules sVCAM-1, sE-Selectin and leptin.Subjects consisted 32 women with physiological menopause (FM) had notherapy,54 women with natural menopause on hormone replacementtherapy (HRT; E2 plus progestin), 26 women with surgical menopause,before estrogen replacement therapy (ERT;E2). Thirty healthy women(had no menopause) served as a control group (K). Serum E2, FSA andLH levels, and also serum glucose, BUN, CRE, AST, ALT, CHOL, HDL, LDL,69and TG levels were evaluated. In each post-menopausal group the meanE2 level was found as <50 pg/ml. In the same groups, the mean valueofFSH was found <30 mIU/ml. The differences between CHOL, HDL, LDL,and TG levels were not found . As a result of our research, we couldn?dfind any difference between the groups mean CAT activity. MDAconcentrations were found significantly lower as compared each of post-menopausal group. GSH concentration was found lower in K group ascompared other groups. As the same, GSH-Px and GR enzymes were fondlower in K as compared with the other groups. In post-menopausal groupsthe mean NO levels were found higher as compared with K. There was nodifference between MPO levels. sVCAM-1 and sE-selectin levels wereevaluated and was found that there was no difference between groups.Also, we evaluated the influence of ERT and HRT, and BMI (kg/m2) on theserum leptin concentration. We found that, leptin concentrations aresignificantly higher in obese post-menopausal women than in their non-obese counterparts. Also, we showed that serum leptin concentrationswere influenced by ERT end HRT concentrations. Taken together, it isobvious that menopause was disturbed oxidative system and lipidperoxidation, and influenced metabolism of GSH. In addition, we couldn?tfind any differences according to the parameters in ERT end HRT regimen.70
