Abstract (EN)

Cancer is one of the disease groups with the highest rate of death compared to other diseases. The biggest problem in such disorders is the rapid growth and proliferation of the cells and the threat of metastasis to various tissues. Since the side effects of the drugs and techniques applied to cure cancer patients are very severe, a new OZF2 phenothiazine derivative was synthesized in our study as a stronger and more effective anticancer drug candidate. Phenothiazine derivatives are frequently used in both medicine and industrial fields. Although the biological activities of these derivatives are quite wide, they are among the drugs preferred in many disease groups. After synthesizing the N-substituted phenothiazine derivative as part of this study, its molecular structure was confirmed by 1H NMR and 13C NMR. The molecule contains electronegative oxygen and chlorine atoms in the long alkyl chain attached to the nitrogen atom in the phenothiazine ring. The anti-cancer activity of the OZF2 phenothiazine derivative was tested with the MTT test on the colon cancer cell line, which is among the cancer types with the highest incidence worldwide. The MTT test measures cell viability by conversion of tetrazolium salt (3(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) to purple formazan. In our study, OZF2 phenothiazine derivative at various concentrations (1,2,4,8 and 10 ug/ml) was incubated with the colon cancer cells for 24-48-72 hours. Then, we incubated the phenothiazine derivative at different concentrations (0.5-1-3 ug/ml) for 24 hours with macrophage cells. Anti-inflammatory activity was tested by evaluating the release levels of pro-inflammatory cytokines TNF-α and IL-6 cytokines in macrophage cells treated and untreated with LPS.
