Epigenetic analysis of candidate tumor suppressor genes in papillary renal cell carcinoma

Abstract (EN)
Papillary renal cell carcinoma (pRCC) accounts for 10-15% of renal cell carcinomas. pRCC is a type of renal cell cancer resistant to classical chemotherapy and immunotherapy. Altough the role of DNA hypermethylation in pathogenesis of RCC was demonstrated previously, our knowledge in pRCC in limited. The aim of this study was to determine the methylation status of CST6, RPRM and GREM1 genes in pRCC patients. After bisulfite conversion of DNAs isolated from paraffine embedded tumor and adiacent normal tissues of 22 patients. Methylation Spesific PCR was performed. Methylation frequencies for GREM ,CST6 and RPRM genes were found as % 41 (9/22) ,14% (3/22) and 59% (13/22), respectively. No significant association was found between methylation status and sex, age and pathological grade (p>0.05). As a result; it can be thought that promoter methylation in GREM1,CST6 and RPRM genes may have a role in the pathogenesis of pRCC. However, further studies by using different molecular techniques are needed to clarify these results.
