Abstract (EN)

In the present work, experimental design was used in tablets that, ıncluding paracetamol, caffeine and propiphenazone by using HPLC method and this method validated. İnvestigated factors which are, pH, flow rate and methanol ratio in mobile phase in optimisation of experimental design. This factors and interactions between factors were researched on effect of resolution, peaks in choromatogram. 23 factorial design was used for it. Two level were chose for each factor. This levels are 2,8 and 3,2 for pH, 0,8 and 1,5mldak-1 for flow rate, 250mL / 1000 mL and 355mL / 1000 mL for methanol ratio. 24 experiments were done totally. Order of experiments were determinated at random. Low level of factors were showed as (-), high level were showed as (+). ANOVA applied investigated factors and interaction between factors of effect on resolution. The results showed that all factors have effect on resolution. But between thr factors, flow rate is more effective others. Methanol ratio is more less effective than flow rate . pH is less effective on resolution. On validation works, averages recoveries of paracetamol, caffeine and propiphenazone were found to be 101,07 % ; 97,00 % ; 100,60 % Relative standart deviation (RSD) values of within day analyses were in range of % 1,61 ± 0,42 ; %1,12 ± 0,28; %1,69 ± 0,73 for paracetamol, caffeine and propiphenazone, respectively. Meanwhile relative standart deviation values of day-to-day analyses were found to be %0,56± 0,07; %3,84 ± 0,93; %1,68 ± 0,15 for paracetamol, caffeine and propiphenazone, respectively. The main materials of concentration were prepared in solvent of analyses equivalent of 50% -150 % concentration in range , corelation coefficients calculated 0,9986;0,9979;0,9985 for paracetamol, caffeine and propiphenazone, respectively.
