Effect of plant regulators and medium culture on in vitro micropropagation of Portugal Gandera local sweet potato (Ipomoea batatas (L.) Lam) genotype

Abstract (EN)
This work accomplished at the micropropagation laboratory at ESAC/IPC in 2019/2020. Different culture media prepared to test the in vitro micropropagation of a local genotype of sweet potato (Ipomoea batatas (L.) Lam.) used at the Gandera region of Portugal. Objective of this study the effect of plant regulators added to three different medium culture, on in vitro propagation of a regional genotype of sweet potato (Gandera) cultivated at sandy soils. To these media culture, it is added different regulators as Benzyladenine (BA), Gibberellic acid (GA3) and 1-Naphthalene acetic acid (NAA), at different concentrations singly or in combination. To evaluate the effect of the media culture and plants regulators on multiplication of in vitro cultures the following parameters recorded: number of shoots, length of shoots, degree of callus, number of roots, length of roots, plant weight and root/shoot rate. Number of shoots observed 2.95±0.26 in media culture D and 3.00±0.53 in MS control. A higher number of roots was observed (2.25±0.45) at the medium culture control MS. These results showed that MS control were observed consequently with a higher weight (0.150±0.03 g). The addition of NAA to the media culture increased the presence of callus and reduced the shoot length. This study results showed that a higher number of shoots was observed at the media culture D (½MS) which supplemented with BA, GA3 and NAA, at the following concentrations 0.5, 2.0 and 0.05 mg/l, respectively and MS control. It is consider that the results obtained from this study can be used in in vitro micro propagation of sweet potato varieties and other sweet potato genotypes.
