Isolation and İdentification of Bifidobacteria can be Used as Probiotic Bacteria Probiotics can be defined as living microorganisms of which has proved beneficial effects on health of the host and that modulate the intestinal flora. Besides, it has been stated that they threats or protects against many illness. İnterests on the subject and consumption of these products are presumeably increasing. From fermantation to biological processes, from agricultural processes to foods and especially recently in medicine area, Lactic Acid Bacteria (LAB) have became a significant issue of researches and researches on LAB have been zoomed in. On this work breast milk and breastfed infants feces were decided to use as source and Bifidobacterium strains, which are LABs and have probiotic speciality, are isolated using TPY agar modified by the addition of mupirocin (50mg/L) and charecterized be using fructose-6-phosphate phosphoketolase enzyme activity becide testing with classical bio chemical methods. Some important properties in terms of probiotic use such as resistance to acid (pH 3.0, 4.0, 5.0, 6.0, 7.0 ve 8.0) and antimicrobial activities of these isolates has been examined. Looking at the results of studies on acid tolerance has been a significant decrease in bacterial concentration when working in more acidic conditions (pH 3.0-4.0). However, it is seen that some of the bacteria retain their viability pH 3.0 value. pH 5.0 and pH 6.0 has also been an increase in the number of bacteria observed pH 7.0-8.0 maximum development. Antimicrobial activity of the isolated bacteria of the study have showed high antimicrobial activities on tested bacteria E. coli ATCC 25922, S. aureus ATCC 6538 P.
