Abstract (EN)

Polyhydroxyalcanoates (PHA) might be cite to biodegradable plastics which have same physical and chemical feature with plastics. Polyhydroxyalcanoates (PHA) are polyesters of hydroxyalcanoates (HA). Although there is enough knowledge extracellular degradation of PHA which is produced under the extreme amount of carbon source and limitted amount of essential nutrient (like N, P, S, O ve Mg) condition by microorganism, there is not enough knowledge about determination of intracellular degradation pathways. Within the scope of the thesis, Pseudomonas aureofaciens is selected as a model. By altering the C/N ratios of culture medium the microorganism is primarily induced for PHA synthesis and degradation pathways and then organic acids which are intracellular PHA degradation are determined. PHA profiles are estimated in induced PHA producing, starving and non-induce PHA producing conditions. As result of analysis following findings were determined: formic acid concentration differs expecially in starving and non-induce PHA producing conditions, valeric acid concentration is higher than other organic acid in induced PHA producing, starving and non-induce PHA producing conditions, isobutyric acid only is determined in induced PHA producing and glutamic acid, threonin, valine amino acids concentrations differs from other amino acid concentrations farklılık göstermesi bu organik asitlerin PHA metabolizması ile ilişkili olduğunu göstermektedir. All of them show that these organic acids can be related with PHA metabolism. All of these results which will be precursor of new research related with PHA degradation metabolic pathway, will shed light on this researches.
