Abstract (EN)

Objective: The aim of this thesis is to clone the gene encoding the phytase enzyme of the bacteria Pseudomonas Putida used as a biofertilizer. Material and Methods: The phytase gene of the P. putida KT2440 strain was examined in bioinformatics and the phytase gene was determined with the obtained genome information, and a primer belonging to the promoter and structural gene regions was designed for the cloning of this gene. The phytase gene of P. putida KT2440 strain was cloned with the obtained primers and mutant stock cultures were created. Result: Within the scope of the thesis study, targeted gene regions of P. putida KT2440 strain were amplified by PCR method and transferred to the vector. Recombinant vectors were transformed into compotent E. coli cells. Recombinant DNA fragments were detected in the screening results and cultures of cells carrying these fragments were created. Conclusion: Rekombinant cultures were obtained from the strains obtained by transforming the DNA region of the phytase gene into recombinant cell lines. It seems possible to use these clones in future studies on the phytase gene and phytase enzyme.
