ABSTRACTMaster ThesisINVESIGATION OF ANTIOXIDANT PROPERTIES SAPONINECONTAINING PLANTS AGAINST RADIATIONÖmer YALINKILIÇUniversity of Afyonkarahisar Kocatepe Graduate School of Natural and Applied SciencesDepartment of Chemistry, Supervisor: Asst. Prof. Dr. Hüseyin ENGİNARProtective effect of saponin containing extracts from three plant species used inTurkish traditional medicine against X-radiation (XR)-induced oxidative stress in ratwere evaluated for in lipid peroxidation (LPO) product and levels of glutathione:Aesculuc hippocastanum L. seed extract (AKE), Medicago sativa L. extract (YE) andSpinacia oleracea L. extract(IE). We assayed the effects of XR dose (5 Gy, 10 Gy) andXR+administration of plant extracts(25.0 mg/kg, 50.0 mg/kg) on serummalondialdehyde (MDA), glutathione (GSH), reduced ascorbic acid (AA), retinol andβ-caratone concentrations in rats. Only giving XR groups, the plasma MDA (p <0.001)value significantly increased but GSH (p <0.01), AA, retinol and β-caratoneconcentration in blood decreased with increasing XR doses. In the XR+extracts-treatedgroups, plasma concentrations of MDA were increased significantly with increasingradiation but their concentrations were decreased significantly with increasing extractconcentrations. Plasma concentrations of GSH, β-carotene, retinol and AA inXR+extracts-treated groups were decreased importantly with increasing XR but theirvalues increased with increasing extract concentrations. And also blood samples ofXR+extracts-treated groups compared with control group, GSH, β-carotene, retinol andAA significantly increased but only MDA values significantly decreased. The resultsshowed that all extract have enhanced the antioxidant status and have decreased theincidence of free radical-induced lipid peroxidation in blood sample of rats exposed XR.However, antioxidant effect of AKE given animal was more effective than that of YEand IE administered whole-body XR rats.2007, 64 pagesKeywords: Aesculuc hippocastanum L. seed extract, Medicago sativa L. extract,Spinacia oleracea L. extract, Lipid peroxidation, X-rays, Rat
