Abstract (EN)

The urinary bladder has two functions as storage and discharge of urine. The storage of urine is achieved by a functional sphincter and the discharge of urine is achieved by a functional detrusor formed as a web with smooth muscle fibers. The urinary bladder interior pressure increases and urine discharges by the contraction of detrusor and interior pressure decreases and urine storage occurs by the relaxation of detrusor. The fullness sensation during the storage of urinary bladder is perceived by receptors placed inside the muscle layers of bladder walls and these signals is transmitted by afferent nerves. Out of these receptors, α adrenergic and cholinergic receptors plays role in contraction and β adrenergic receptors in relaxation. Some diseases cause neurogenic lower urinary tract dysfunctions by affecting receptors and neurosystem and drugs used in case of neurogenic lower urinary tract dysfunctions for treatment and mentioned in literature leads to overactive or underactive bladder conditions opened the way for us to investigate the response of precontracted bladder smooth muscles with ACh by applying SG-Benz, Atorvastatine Ca, Caffeic Acid Phenil Ester, Ammonium Pyrrolidine Dithiocarbamate. The study is operated in four groups. Each Cape, Atorvastatine Calcium, Pyrrolidine and SG-Benz formed a group with 10 animals and groups total of 40 animals. The rat used in the experiments were weighted first and then killed by cervical dyslocation and the abdominal part is removed by medial line incision in the interior of ureter of bladder. The isolated bladders were fixed to computerized organ bath systems filled with Krebs-Henseleit solution. The organ vitality is tested by KCl and the ACh was given to bladder for precontraction. CAPE, Atorvastatin calcium, pyrrolidine or SG-Benz were given as 10-9 - 3x10-4 M doses cumulatively to obtain contraction and relaxation responses and these responses were recorded in data acquisition analyzing system by isometric transducer. There are meaningful differences at definite doses among groups and the highest relaxation response is found in the samples provided with SG-Benz. The shared response dose of relaxation responses of all drugs was detected as 3x10-6 M. The SG-Benz at this dose produced more relaxation response than all other drugs used. Then, pyrrolidine, atorvastatine and CAPE come respectively. As a result, all drugs produced dosage related increasing relaxation responses in precontracted bladder smooth muscle preparations with ACh applied together.
