Abstract (EN)

June 2023, 99 Pages Diabetes is an endocrine and metabolic disease that causes damage to kidney tissue as well as to all tissues. It is thought that probiotics can be effective in the treatment and prevention of diseases due to their health benefits when taken in sufficient quantities. In our study, it was aimed to investigate the effects of probiotics in diabetes. In our study, 34 male Wistar albino rats were used and they were divided into 5 groups as group K, group P, group Dm, group DmP and group PDm. Group K rats were administered PBS by oral gavage for 8 weeks, and 0.6 mg/kg VSL#3 probiotic supplement was administered to group P rats by oral gavage for 8 weeks. Diabetes was formed by administering a single intraperitoneal dose of 50 mg/kg streptozocin to Group Dm. The rats in Group PDm were given probiotic supplement during the first 4 weeks of the study, then diabetes was established, and probiotic administration was continued for 4 weeks after diabetes was established. Diabetes was induced in rats in Group DmP with streptozocin (50 mg/kg intraperitoneally), and then probiotics were administered for 4 weeks. At the beginning and end of the experiment, blood glucose measurements of the rats were made, and the blood glucose value was accepted as 250 mg/dl for the rats in which diabetes was desired. At the end of the experiment, the rats were sacrificed under anesthesia by administering 90 mg/kg ketamine + 10 mg/kg xylazine intraperitoneally. Blood serum samples were obtained for biochemical evaluation and kidney tissues for histochemical analysis. As a result, we observed that probiotics have a protective effect on blood glucose levels against the damage caused by diabetes, but have no therapeutic effect. We concluded that probiotics have a curative effect against the increase in serum urea and BUN caused by diabetes. But we did not see any significant change for creatinine. In our histochemical evaluations, we determined that probiotics have a positive effect on kidney tissue damage caused by diabetes. We also observed the positive effects of probiotics against diabetes by immunohistochemically alpha-SMA and Bcl-2 expressions.
