Abstract (EN)

Interest in the determination of flavonols concentrations in medicine plant and vegetables, including myricetin, quercetin and kaempferol which have anticancerogen properties, is increasing in recent years. Particularly, widespread usage of HPLC-MS system that is combining to very good separation method-HPLC and both sensitive and reliable detector-MS, make their determinations possible for more kinds of foods.In this study, the parameters that may affect the signal of myricetin in the HPLC-MS were optimized. The mixture of water/methanol/CAN/formic acid was used as mobile phase. The optimum parameters were found to be injection volume=5.0 mikro liter, flow rate of mobile phase=0.6-0.8 ml/min, column temperature=30-40 oC and fragmentor=140-170 V. The optimum parameters observed were applied to determination of flavonols in fruits and vegetables grown in Elazig vicinity. So, determination of myricetin, quercetin and kaemferol in fruit and vegetabels such as terebinth (Terebinthina chica L.), nettle (Urtica dioica L.), rosa hip (Rosa canina L.) and purslane (Portulace oleracea L.) by HPLC-MS were carried out. Flavonols in plant samples were extracted by mixture of methanol/ascorbic acid/HCl. From the obtained results, the highest and the lowest concentrations for myricetin, quercetin and kaemferol were found to be Portulace oleracea L. (47 ppm) and Rosa canina L. (0.63 ppm), Portulace oleracea (39 ppm) and Terebinthina chica (< t.s), Urtica dioica (7.7 ppm) and Terebinthina chica (< t.s), respectively.
