Determination of cellular antioxidant and antiproliferation activities of Sideritis perfoliata extracts on HeLa cells

Abstract (EN)

In this study, we aimed to determine anticancer and antioxidant activites of methanol extact of Sideritis perfoliata (SPM). For this aim, doses in different concentration of SPM were applied to determine the cell viability of HeLa cells by MTT method. After administration of SPM, apoptosis was determined by Annexine V and propodium iodide staining. Cytokines IL6 and IL8 levels were determined in supernatants which taken from tubes. Antioxidant activity of SPM was observed with DPPH (1,1-difenil-2-pikrilhidrazil) and DNAnicking test systems. Some of phytochemicals of SPM were determined by LC-MS-MS. SPM inhibitedproliferation and induced apoptosis in HeLa cells. Although IL6 and IL8 levels were increased in doses 25,50, 100 µg/ml, they were reduced in dose 200 µg/ml of SPM. SPM showed the DPPH (1,1-difenil-2-pikrilhidrazil) scavenging activity in a dose dependent manner also. DNA protection activity was observed in all of the doses. Quercetine, acetohydroxycinnamic acid, vanillic acid, resvaratrol, caffeic acid, acetohydroxycinnamic acid, alizarinand salicylic acid were determined in SPM. As a result, it can be said that SPM has an anticancer activition HeLa cervical cancer cells through induction of apoptosis, and is a source of natural antioxidants.
